According to People, was born in May 1998 to a prominent family outside , Maryland. His late grandfather was a multimillionaire real estate developer and his cousin, Nino Mangione, is a member of the Maryland House of Delegates, according to .

Mangione attended an all-boys private school in Baltimore called Gilman School, where he graduated as valedictorian in 2016, as per People. He then studied engineering at the University of Pennsylvania and graduated in 2020. After college, he moved to Hawaii but stopped communicating with his family around June 2024.

Mangione has two sisters, MariaSanta and Luciana. MariaSanta is a physician, while Luciana is a visual artist.

On the night Mangione was arrested, his cousin Nino shared a statement from the family, saying they “cannot comment on news reports regarding Luigi Mangione” and that they only knew what they had “read in the media.” He added that the family was “shocked” and “devastated,” and offered prayers for Brian Thompson’s family, as per .
